Hemoglobin A2 Quik Column Assays
Test used to assay Hb A2:
Anion exchange microhematography
Principle of test:
A hemolysate of patient’s red cells is added to top of resin column containing cellulose attached to positively charged molecules. The hemoglobins are selectively bound to positive molecules based on pH of buffer added. Hb A2 passes through the column and is collected. The amount of hemoglobin A2 is then determined and compared to patient’s total hemoglobin and % Hb A2 is calculated.
Elevated Hb A2:
b-thalassemia heterozygous or homozygous
Decreased Hb A2:
Iron deficiency anemia, Hb H disease, sideroblastic anemia
