Ascorbate-Cyanide Test Test
Principle:
Whole blood is added to sodium cyanide and sodium ascorbate reagent. H2O2 is produced by reaction between hemoglobin and sodium ascorbate. Sodium cyanide blocks red cell catalase from reducing H2O2. If the H2O2 is not reduced by the hexose monophosphate shunt the hemoglobin will be reduced forming a brown substance.
Clinical application:
Glucose-6-Phosphate dehydrogenase deficiency:
A positive test result (brown color) will be obtained
