Direct Antiglobulin Test (DAT)
Principle:
Erythrocytes with antibodies attached will agglutinate when antihuman globulin is added to washed cell suspension.
Clinical application:
 antibody sensitized red cells
return
Procedure:
 3-5% suspension of patient’s red cells are washed three times
 2 drops of antihuman globulin added, mixed, and centrifuged
 as cells are resuspended they are checked for agglutination
