When
RBCs containing HbS are mixed with a reducing substance
such as sodium metabisulfite the decreased oxygen tension will
cause the red cells to sickle.
1. Blood is placed on slide and
sodium metabisulfite
is added

2. A coverslip is placed over blood
and sealed
3. Blood is examined for presence of sickle cells
